چکیده

Purpose We investigated the extracellular matrix (ECM) of the lamina cribrosa (LC) and peripapillary sclera (PPS) and compared histone acetylation and related enzymes to identify racial differences between Korean and Caucasian donor eyes. Methods Posterior segment tissues were obtained from 30 Caucasian donors and 42 age and axial length-matched Korean donors. Histone modification was assessed for histone deacetylase (HDAC) 2, HDAC3, and acetylated histone H3. The promoter regions of the major ECM in the LC and PPS including collagen type I and III, and elastic fiber components (elastin and fibrillin-1) and lysyl oxidase enzymes including lysyl oxidase-like 1 and 2 (LOXL2) were evaluated by chromatin immunoprecipitation (ChIP) assay. Protein and mRNA expression of major ECM components were assessed using real-time polymerase chain reaction analysis, western blot analysis, and immunohistochemical staining. Results HDAC2 and HDAC3 expression levels were decreased and acetylated histone H3 was increased in the LC and PPS of Korean eyes than Caucasian eyes. The promoter regions of LOXL2, elastin, and fribrillin-1 genes were highly acetylated in Korean LC. Expression of LOXL2 and elastic fiber components (elastin and fibrillin-1) were significantly increased in Korean LC and PPS than Caucasians according to the real-time polymerase chain reaction, western blot analyses, and quantification of elastic fiber staining. Conclusions Histone acetylation status differed in the promoter regions of the elastic fiber components and LOXL2 in the LC and PPS according to race. Further study to reveal the association with these findings to the pathogenesis of glaucoma in Korean eyes is needed.

Cell adhesion glycoproteins in the human lamina cribrosa.

PURPOSE The distribution of the cell adhesion glycoproteins, laminin, fibronectin, tenascin, vitronectin, thrombospondin, and entactin/nidogen, was examined in the human lamina cribrosa. METHODS Frozen sections of the optic nerve head from 7 normal human elderly donors were stained by immunohistochemistry. RESULTS All six glycoproteins were detected in this tissue. While laminin and entacti...

3-D histomorphometry of the normal and early glaucomatous monkey optic nerve head: lamina cribrosa and peripapillary scleral position and thickness.

PURPOSE To delineate three-dimensionally the anterior and posterior surfaces of the lamina cribrosa, scleral flange, and peripapillary sclera, to determine the position and thickness of these structures within digital three-dimensional (3-D) reconstructions of the monkey optic nerve head (ONH). METHODS The trephinated ONH and peripapillary sclera from both eyes of three monkeys with early gla...

Intraocular Pressure Effects on Collagen Crimp in the Peripapillary Sclera

INTRODUCTION Glaucoma is the second leading cause of irreversible blindness worldwide with neural damage starting within the lamina cribrosa of the optic nerve head (ONH). It is known that elevated intra-ocular pressure (IOP) is the main risk factor for glaucoma, and many studies have shown that this is because glaucoma is related to a biomechanical imbalance in the eye [1]. Studies have also s...

Variation in the Three-Dimensional Histomorphometry of the Normal Human Optic Nerve Head With Age and Race: Lamina Cribrosa and Peripapillary Scleral Thickness and Position

Purpose This study quantified the thickness and depth of the lamina cribrosa (LC) and peripapillary scleral thickness in high-resolution three-dimensional (3D) fluorescent reconstructions of the optic nerve head (ONH) in eyes from donors of African (AD) and European descent (ED). Methods A total of 64 eyes (45 ED, 19 AD) from 51 normal donors were obtained within 6 hours of death and fixed at...

Central corneal thickness and thickness of the lamina cribrosa in human eyes.

PURPOSE Since central corneal thickness may inversely influence the amount and rate of progression of glaucomatous optic nerve damage and because lamina cribrosa thickness may be of importance in susceptibility to glaucoma, it was the purpose of the present study to evaluate whether central corneal thickness is related to lamina cribrosa thickness. METHODS The histomorphometric study included...
